Suspend 46 gms. in 1000ml distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15lbs pressure (121°C) for 15 minutes. Cool below 50°C. Before gelling, aseptically add sterile defibrinated blood to a final concentration of 5% v/v in B616 and add rehydrated contents of 1 vial of McBride Listeria Supplement to B616/B636. Mix well and pour into sterile petri plates. |
