Suspend 45 gms. in 1000ml. distilled water. Heat to boiling to dissolve the medium completely. Dispense in test tubes and sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Cool the tubes medium is slanted position. Just before inoculation add 0.15 ml. or 2 drops of 50% dextrose solution to each slant. |
