Suspend 51 gms. in 1000 ml. distilled water. Boil to dissolve the medium completely. Sterilize by autoclaving at 15lbs pressure (121°C) for 15 minutes. When to be enriched with haemoglobin, suspend 10.2 gms. in 100 ml distilled water. Sterilize as above. Cool to 50°C and aseptically add 100 ml of 2% sterile haemoglobin solution.Mix well and pour into sterile petri plates. |
